1.  Single binding site: Assume only one ligand binds to each receptor

Usually in an experiment [L] is varied and measures [B].  The desire is to estimate KD (or its inverse KA = 1/KD) and [RT].  If you know the total receptor protein molar concentration [P], then [RT]/[P] = number of binding sites/molecule of protein.
